Influenza virus A gives rise to influenza in birds and mammals, including humans. Some strains of influenza A virus can cause severe diseases and be potentially fatal if left untreated or may lead to human influenza pandemics due to its highly infectious nature. When an influenza A virus infection occurs, the human immune system produces specific antibodies to destroy the invading virus. Thus, this seroconversion allows the person to have antibodies for the pathogen after the infection. To determine if one has been exposed to certain infectious agents, a serological assay which measures the levels of pathogen-specific antibody in the blood can be used. In this practical, we will explore the use of indirect immunofluorescence antibody assay (IFA) in monitoring the presence and localization of influenza antibodies. The stained cells are then analysed by fluorescence microscopy.
