Atherosclerosis causes coronary heart diseases by depositing fats and cholesterol in the arterial walls, thus causing blockage in the blood vessel. Today, coronary heart disease is one of the leading causes of deaths around the world as our diets and exercise regimes undergo various changes. Coronary heart disease can be prevented if early genotype testing can allow people to know if they are at greater risk of the disease and take adequate measures to adjust their diets and incorporate exercise. This can be done by exploring the levels of Cholesteryl Ester Transfer Protein (CETP) and thus, the CETP gene which changes the levels of lipoproteins can be further examined by Restriction Fragment Length Polymorphism (RFLP) analysis. In this practical, we examine CETP Taq 1B polymorphisms by amplifying DNA fragments encoding the CETP gene by Polymerase Chain Reaction (PCR) amplification, followed by restriction digest with Taq 1 enzyme and gel electrophoresis to separate and visualise the bands. By analyzing the bands on the agarose gel, we are able to determine the genotype of the samples and thus, evaluate if the persons are prediposed to cardiovascular dieases, and even obesity and Type 2 diabetes.
