Transformation of cells is widely used in genetic engineering. It plays a vital role in the development of molecular biology. The main purpose of implementing this technique is to introduce a foreign plasmid into bacteria (The American Phytopathological Society, n.d.). Then, the plasmid can use the bacteria as a medium to amplifies and form larger quantities (The American Phytopathological Society, n.d.). There is a total of 4 key procedures in bacterial transformation which are the preparation of competent cells, the transformation process, the cell recovery period and the final cell plating process (Thermo Fisher Scientific, n.d.). Firstly, the competent cell preparation must be carried out in order to counter back the low natural competency of E. coli. Next, the process of transformation is then carried out by using a heat shock or electroporation process. Moreover, the cell recovery period means that the transformed cells are cultured and kept in an antibiotic- free medium to enhance the expression of antibiotic resistance gene. Lastly, the cell plating process is carried out to determine the recovery of successful transformants (Thermo Fisher Scientific, n.d.).
